League games - yes (as teams are playing each other home and away)
Regional Cup / AFLC games - no. Let's say that these games are played with no advantage because 50% of the tickets are sold to supporters of one team and 50% to the others. So even if such game is played on your stadium, you do not get home advantage.

And how does the home advantage manifests itself during the game? What is the real 'advantage' of playing at your home?

In the games on "neutral" ground, the ticket income is split 50/50 amongst the two teams

With HA your players get a bit of a "boost" on their skills.
